Des mutations dans le g\u00e8ne LMNA entra\u00eenent des pathologies de s\u00e9v\u00e9rit\u00e9 variable, les laminopathies. La plus s\u00e9v\u00e8re est la prog\u00e9ria, avec le vieillissement pr\u00e9matur\u00e9 de tout l\u2019organisme. Les laminopathies ont des ph\u00e9notypes communs comme des noyaux de forme anormale ou un vieillissement cellulaire pr\u00e9matur\u00e9 (dit s\u00e9nescence). Les principales \u00e9tudes ont port\u00e9 sur prog\u00e9ria et montr\u00e9 que les noyaux sont plus rigides. On ignore si c\u2019est sp\u00e9cifique ou commun \u00e0 toutes les laminopathies. La relation entre mutations, alt\u00e9rations m\u00e9caniques des cellules et s\u00e9v\u00e9rit\u00e9 de la maladie est encore incomprise, ce qui emp\u00eache le d\u00e9veloppement d\u2019outils de diagnostic et th\u00e9rapie.<\/span><span style=\"font-family: Calibri,Helvetica,sans-serif,EmojiFont,Apple Color Emoji,Segoe UI Emoji,NotoColorEmoji,Segoe UI Symbol,Android Emoji,EmojiSymbols; font-size: small;\"><br \/>\n<\/span><span style=\"font-family: Calibri,Helvetica,sans-serif,EmojiFont,Apple Color Emoji,Segoe UI Emoji,NotoColorEmoji,Segoe UI Symbol,Android Emoji,EmojiSymbols; font-size: small;\"><br \/>\nMa th\u00e8se visait \u00e0 \u00e9tudier cette relation \u00e0 l\u2019\u00e9chelle de la cellule et sur des temps courts (secs). J\u2019ai d\u00e9velopp\u00e9 un syst\u00e8me microfluidique pour suivre la d\u00e9formation de cellules dans des constrictions. Nous avons combin\u00e9 la microfluidique avec de l\u2019analyse d\u2019image semi-automatis\u00e9e et un mod\u00e8le rh\u00e9ologique pour extraire les propri\u00e9t\u00e9s m\u00e9caniques des cellules.<\/span><span style=\"font-family: Calibri,Helvetica,sans-serif,EmojiFont,Apple Color Emoji,Segoe UI Emoji,NotoColorEmoji,Segoe UI Symbol,Android Emoji,EmojiSymbols; font-size: small;\"><br \/>\n<\/span><span style=\"font-family: Calibri,Helvetica,sans-serif,EmojiFont,Apple Color Emoji,Segoe UI Emoji,NotoColorEmoji,Segoe UI Symbol,Android Emoji,EmojiSymbols; font-size: small;\"><br \/>\nNous avons \u00e9tudi\u00e9 des fibroblastes pr\u00e9matur\u00e9ment s\u00e9nescents de patients (mutation R482W) et des fibroblastes d\u2019un individu sain trait\u00e9s avec un inhibiteur de prot\u00e9ase qui alt\u00e8re la lamine A\/C. Ces cellules ont une viscosit\u00e9 plus grande qui domine leur dynamique de d\u00e9formation.
